  • Background: Antioxidants are chemical compounds that can donate one or more electrons to free radicals to prevent oxidation reactions. One part of the plant that has potential as natural antioxidants is Jatropha gossypifolia Linn barks which contain secondary metabolites flavonoids and polyphenols. The activity of plant depends on the selection method of extraction. Kinetic remaceration is one method that can extract optimally of antioxidant compound from the bark of Jatropha gossypifolia Linn. Aim: To determine the reduction ability of ethanol extract of Jatropha gossypifolia Linn barks which extracted by two methods, non-kinetic and kinetic remaceration against free radical DPPH and comparing the antioxidant activity from both methods. Methods: In vitro DPPH. Jatropha gossypifolia Linn barks extracted by two methods, non-kinetic and kinetic remaceration using ethanol 96% as a solvent. Then to identify the chemical compounds profile by TLC method. Measurement of antioxidant activity aims to determine the IC50 value of sample using DPPH method quantitatively by UV-Vis spectrophotometer. Results and Conclusion: The result of chemical compounds profile by TLC showed that ethanol extract of Jatropha gossypifolia Linn barks is positive contains alkaloids, terpenoids, flavonoids, polyphenols, and anthraquinone. The IC50 value of non-kinetic and kinetic remaceration were found to be 8.47 Âμg/ml and 8.09 Âμg/ml. IC50 value of vitamin C as a positive control were found to be 1.47 Âμg/ml. The result of statistical analysis by SPSS using t-Test Independent can be concluded that both methods have a similiar scavenging capability against free radical DPPH (p = 0.684 >α = 0.05).
