ISOLASI DAN UJI POTENSI BAKTERI ENDOFIT SEBAGAI PENAMBAT NITROGEN DAN PENGHASIL HORMON IAA (Indole Acetic Acid) DARI EMPAT JENIS TANAMAN PADI (Oryza sativa)

  • Endophytic bacteria can live anywhere in plant tissue and as we know they can contribute for nitrogen fixation and produce plant hormones for host plant. This research are done to get some isolate of endophytic bacteria that are potential for nitrogen fixation and indoleacetic acid production from different kinds of rice, such as Beras Merah, Mentik Wangi, Beras Hitam, and Ketan Merah. Bacteria was isolated from rice plant tissue and was pured with streak plate method in Nutrient Agar (NA) medium. Nitrogen fixation test, the bacteria isolate was growth in modified Nitrogen Free (NF) medium and the number of bacterial population in the culture was read in UV-spectrophotometer at 420 nm absorbance. IAA production were tested by inoculated bacteria isolates to M63 + trypthopan medium. The amount of IAA produced per milimeter culture was estimated using a standard curve. The result showed that thirty two endophytic bacteria isolates have the potential for nitrogen fixation and able to produced indoleacetic acid hormones. The characterization of bacterial colony was dominated with circular form, white-grey colour with size 1-7 milimeter and convex surface. There are four endophytic bacteria, the best one from each kind of rice; IM-3, IM-11, IM-21, and IM-32, that showed the best potential for nitrogen fixation. In IAA production test, isolate IM-1 showed the best ability to produced IAA until 7,78 ppm, and followed by isolate IM-10 about 6,62 ppm, isolate IM-25 about 6,31 ppm, and the last is isolate IM-23 about 5,77 ppm.
