- This research was aimed to detect Tobacco Mosaic Viruses (TMV) utilize N gene of tobacco plant resistant using specific primer trough Polymerase Chain Reaction (PCR) analysis technique. This research has been conducted in Biotechnology molecular laboratory University of Muhammadiyah Malang. TMV detection of tobacco has been done by isolation of tobacco DNA which then were amplified using N-gene specific primer. The sequence of this primer were 5’-GCA TCT TCT TCT TCT TCT TC-3’ (Forward primer) and 5’-GAG CCT TTG AGA TTG GCC GC-3’ (Reverse primer), the size of DNA amplified by this primer pairs was about 450 bp. The results of the research showed that N-gene specific primer DNA could be used to detect the existence of TMV on Tobacco. Based on 4 varieties of tobacco used (Paiton, Kemloko, Kasturi and Bojonegoro) which were intentionally infected by viruses and 4 varieties which were not infected by viruses, 7 varieties on them have been detected by DNA viruses marker, except a variety which was not amplified by the primer used was Bojonegoro (Virginia) not infected by TMV viruses.
