  • Tuberculosis (TB) is the most common cause of death in developing countries of which 75% of patients are at productive age of 20-49 years, due to the developing country has a dense population and high prevalence, resulting in more than 65% of cases tuberculosis occurs in Asia. The increased number of cases is due to immune system, nutritional status and personal hygiene, residential dwelling density, and rapid and precise diagnostic tests. The results of the study by comparing the method of examination of Acid Resistant Bacteria microskospik with PCR technique to detect Mycobacterium tuberculosis has a sensitivity of 30%, specificity 80%, and accuracy of 47%. Based on the results of the study, the detection of Mycobacterium tuberculosiswith PCR technique was as good as TB bacterial culture, but the time of PCR technique was shorter than Mycobacterium tuberculosis, and TB bacteria was not detected by microscopic examination (BTA). This study aims to determine the comparison of diagnostic tests of sputum examination with Zhiel Neelsen and PCR (Polymerase Chain Reaction) techniques. The research was conducted in the laboratory of Microbiology Faculty of Medicine and Health Sciences of Syarif Hidayatullah State Islamic University in Jakarta from May to October 2016. The method used was identification of bacteria with Zhiel Neelsen and PCR staining, sputum samples were performed during - in the morning - while (SPS) , the sample came from Bojonggede Public Health Center. Based on the result of the research, the samples were from sputum samples of Bojonggede Puskesmas with 68 samples, the result showed 14 positive samples of BTA and smear positive percentage of BTA by 21%, while negative BTA was 54 samples and the percentage was 79%. Comparison of PCR test result and smearing of BTA as many as 19 samples was the result of smear smear test showed the result of smear positive smear as much as 9 samples and the percentage of 47%, while the negative BTA by 10 samples and the percentage of 53%. PCR test results showed positive PCR test results of 11 samples and a positive percentage of 59%, while negative PCR test results were 8 samples, and the percentage was 42%. The conclusion of this research is positive smear positive PCR test more than the percentage of BTA stain with Zhiel Neelsen method.
