  • Hypertension is considered as a multifactorial trait resulting from a combination of environmental and genetic factors. Renin-angiotensin system genes are the most important factors for investigation of the cause of hypertension. Inter-ethnic differences in cardiovascular diseases indicate the need to examine the association of AT1R gene polymorphism and hypertension in various population. Recent studies showed that AC genotype and 1166C allele have been associated with higher prevalence of hypertension. There is no data reported so far about the possible effect of A1166C polymorphism of the AT1R gene and hypertension in Indonesian population. This study aims to investigate AT1R gene polymorphism in hypertensive subjects in Javanese population. A cross sectional study was conducted in August 2008-Mart 2009 involving 68 subjects from hypertensive group and 45 subjects from normotensive group. Genotypes based on the polymorphisms (AA/AC/CC) using a PCR-RFLP method. The results showed, the genotype frequencies in hypertensive group were 92.65% AA; 7.35% AC; and normotensive groups were 77.78% AA; 22.22% AC (p=0.024). The allele frequencies in hypertensive were 96.27% A and 3.73% C; normotensive group were 88.89% A and 11.1% C (p=0.028). Frequencies of AA genotype and A1166 allele AT1R gene in hypertensive subject were higher than normotensive subjects. The A1166 allele of the AT1R gene was a predisposing factor in hypertension.Keywords: hypertension, RAA system, AT1R A1166C gene polymorphism
