Influence of Preparation of Inclusion Complex of Ketoprofen-β-Cyclodextrin on Dissolution Rate of Ketoprofen

- Dissolution rate of active ingredient is one of determinant in course of absorption dosage form which given by oral. Method of preparation inclusion complex of determining amount of inclusion of active ingredient and influent of dissolution rate. Solid inclusion complex prepared with using kneaded method, coevaporated method, and co-precipitation method with ratio of molar ketoprofen-b-cyclodextrin (1:1). The dissolution test was carried out using the paddle method at a temperature of 37± 0,5oC and 50 rotation per minutes (rpm) in medium of water pH 7. The ketoprofen concentrations were spectrophotometrically determined at absorption maximum 250 nm of ketoprofen. Percentage of complex ketoprofen with kneaded method, co-evaporation, and co-precipitation were 5.3, 54.8 and 58.7%, respectively. Percentage of dissolve ketoprofen at 90 minute in solid inclusion complex of ketoprofen-b-cyclodextrin with kneaded method, coevaporation, and co-precipitation were 41.0%, 89.8%, and 91.8%, respectively. Co-evaporation and copresipitation method result of inclusion complex of ketoprofen-b-cyclodextrin by ketoprofen which inclusion of more amount compared to kneaded method. The rate dissolution increased on inclusion complex of ketoprofen-b-cyclodextrin with coevaporation and copresipitation method than inclusion complex of ketoprofen-b-cyclodextrin with using kneaded method.Keywords: dissolution rate, kneaded, co-precipitation, coevaporation.
