- Rifampicin and isoniazid is one of combination preparations tablet used in the treatment of tuberculosis. Mean Centering of Ratio Spectra (MCR) is one of the methods that has been developed in ultraviolet spectrophotometry for determination of active substances that is contained in a pharmaceutical preparations. The purpose of this research which is to determine a mixture of rifampicin and isoniazid in preparation tablet by mean centering of ratio spectra ultraviolet spectrophotometry method in a solvent HCl 0.1 N. This research was using Rifampicin 150 mg / Isoniazid 150 mg Generic Tablet and Pro TB 2 Tablet which contains rifampicin 150 mg and isoniazid 150 mg and was determined by MCR ultraviolet spectrophotometry method and was measured at wavelengths 234 nm for rifampicin and 267 nm for isoniazid in a solvent HCl 0.1 N. The calculations were done with software MATLAB R2009a version. The research was continued by testing validation to a method of MCR in the analysis the determination of levels, as accuracy, precision, limit of detection, and limit of quatification. The research results shown that Rifampicin 150 mg / Isoniazid 150 mg Generic Tablet is containing rifampicin (96.56 ± 0.84)% and isoniazid (94.04 ± 1.71)%, meanwhile Pro TB 2 Tablet is containing rifampicin (96.63 ± 0.73)% and isoniazid (99.23 ± 0.85)%. The results of the validation of rifampicin for accuracy 100.52%, precision 1.1% , LOD 1.443 μg/ml, and LOQ 4.8113 μg/ml, meanwhile isoniazid for accuracy 99.39%, precision 0.95%, LOD 0.9669 μg/ml, and LOQ 3.2229 μg/ml. Base on the research results could be concluded that MCR ultraviolet spectrophotometry method can be used to determine contents rifampicin and isoniazid in tablet and fulfill the accuracy, precision, limit of detection, and limit of quantification.
- Rifampisin dan isoniazid merupakan salah satu kombinasi sediaan tablet yang digunakan dalam pengobatan tuberkulosis. Mean Centering of Ratio Spectra (MCR) merupakan salah satu metode yang telah dikembangkan dalam spektrofotometri ultraviolet pada penetapan kadar zat aktif yang terkandung dalam suatu sediaan farmasi. Tujuan penelitian ini yaitu untuk menetapkan kadar campuran rifampisin dan isoniazid dalam sediaan tablet dengan metode spektrofotometri ultraviolet secara Mean Centering of Ratio Spectra (MCR) dengan menggunakan pelarut HCl 0,1 N. Penelitian ini menggunakan sampel Tablet Generik Rifampisin 150 mg / Isoniazid 150 mg dan Tablet Pro TB 2 yang keduanya mengandung rifampisin 150 mg dan isoniazid 150 mg dan dilakukan penetapan kadar secara spektrofotometri ultraviolet secara MCR yang diukur pada panjang gelombang 234 nm untuk rifampisin dan 267 nm untuk isoniazid dalam pelarut HCl 0,1 N. Metode perhitungan dilakukan dengan bantuan software MATLAB versi R2009a. Penelitian dilanjutkan dengan pengujian validasi terhadap metode MCR dalam analisis penetapan kadar, seperti akurasi, presisi, batas deteksi, dan batas kuantitasi. Hasil penelitian menunjukkan bahwa Tablet Generik Rifampisin 150 mg / Isoniazid 150 mg mengandung rifampisin (96,56 ± 0,84)% dan isoniazid (94,04 ± 1,71)%, sedangkan Tablet Pro TB 2 mengandung rifampisin (96,63 ± 0,73)% dan isoniazid (99,23 ± 0,85)%. Hasil uji validasi rifampisin yaitu akurasi 100,52%, presisi 1,1%, LOD 1,443 μg/ml, dan LOQ 4,8113 μg/ml, sedangkan isoniazid yaitu akurasi 99,39%, presisi 0,95%, LOD 0,9669 μg/ml, dan LOQ 3,2229 μg/ml. Berdasarkan hasil penelitian yang dilakukan dapat disimpulkan bahwa metode spektrofotometri secara ultraviolet secara MCR dapat digunakan untuk menetapkan kadar rifampisin dan isoniazid dalam tablet dan memenuhi persyaratan akurasi, presisi, batas deteksi, dan batas kuantitasi.
