Uji Aktivitas Antibakteri Ekstrak Etanol dan Fraksi Daun Kucai (Allium schoenophrasum L.) Terhadap Bakteri Escherichia Coli dan Staphyloccocus Aureus

- Chive (Allium schoenoprasum L.) is known to oroginate from North America and Northen Europe. Chive belongs to the family Alliaceae. The objective of this study was to determine antibacterial activity of ethanol extract and fractions of chive. This research includes simplex charateristization, phytochemical screening, extraction of secondary metabolites by maceration with 96% ethanol, fractination with n-hexane, ethyl acetate and the antibacterial activity was measured by agar diffusion method using paper disk against the Escherichia coli and Staphylococcus aureus in which the inhibition zone diameter of bacterial growth was measured. The result of simplex charateristization gave water content 7.98%, water soluble extract 18.28%, ethanol soluble extract 19.49%, total ash value 8.85%, and acid insoluble ash 0.54%. The result of phytochemical screening of chive showed the presence of alkaloids, flavonoids, glycosides, tannins, saponins and steroids/triterpenoids, the n-hexane fraction exhibited to contain steroids/triterpenoids, ethyl acetate fraction showed the presence of alkaloids, flavonoids, glycosides, tannins and saponins. Antibacterial testing of ethanol extract and fractions inhibited antibacterial activity. Ethanol extract with the concentration of 25 mg/ml inhibited the growth of Escherichia coli and Staphylococcus aureus. N-hexane fraction did not show antibacterial activity against Escherichia coli but with the concentration of 300 mg/ml inhibited Staphylococcus aureus growth. Ethyl acetate fraction with the concentration of 5 mg/ml of Escherichia coli and Staphylococcus aureus. Water fraction with 200mg/ml inhibited both Escherichia coli and Staphylococcus aureus.
- Kucai (Allium schoenoprasum L.) diketahui berasal dari Amerika Utara dan Eropa Utara. Tanaman ini termasuk keluarga Alliaceae. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibakteri ekstrak etanol dan fraksi daun kucai. Penelitian ini meliputi karakterisasi simplisia, skrining fitokimia, ekstraksi dilakukan secara maserasi dengan pelarut etanol 96%, fraksinasi menggunakan pelarut n-heksana dan etil asetat serta uji aktivitas antibakteri dengan metode difusi agar menggunakan kertas pencadang terhadap bakteri Escherichia coli dan Staphylococcus aureus dengan mengukur diameter zona hambat pertumbuhan bakteri.Hasil pemeriksaan karakteristik simplisia diperoleh kadar air 7,98%, kadar sari larut air 18,28%, kadar sari larut etanol 19,49%, kadar abu total 8,85% dan kadar abu tidak larut dalam asam 0,54%. Skrining fitokimia simplisia dan ekstrak etanol menunjukkan adanya golongan senyawa alkaloid, flavonoid, glikosida, tanin, saponin dan steroida/triterpenoida, fraksi n-heksana positif steroid/triterpenoid, fraksi etil asetat positif adanya alkaloid, flavonoid, glikosida, tannin dan saponin. Ekstrak etanol dan fraksi memiliki aktivitas sebagai antibakteri. Ekstrak etanol memberikan nilai KHM 25 mg/ml pada bakteri Escherichia coli dan Staphylococcus aureus. Fraksi n-heksana tidak memberikan daya hambat pada Escherichia coli tetapi memberikan nilai KHM 300 mg/ml pada bakteri Sthapylococcus aureus. Fraksi etil asetat memberikan nilai KHM 5 mg/ml pada bakteri Escherichia coli dan Staphylococcus aureus. Fraksi sisa memberikan nilai KHM 200 mg/ml pada bakteri Escherichia coli dan Staphylococcus aureus.
