- Immunomodulator is a substance or drug that can modulate the function and activities of immune system either with stimulating or repairing the function. Immunomodulator can be obtained from plants, one of which is African Leaves (Vernonia amygdalina Delile) that was reported to have function to increase immune system including flavonoids, glycosides, and saponins. The purpose of this study was to investigate the immunomodulatory effect of ethanol extract of African Leaves on hypersensitivity response and antibody titer of immune cells of male mice. This study was conducted by experimental with hypersensitivity response test and antibody titer of immune cells of male mice test. Hypersensitivity response test was evaluated based on the swelling of the feet of mice, while antibody titer test was evaluated by hemaglutination. Twenty five male mice were divided into five groups that is vehicle control group was given CMC Na 0.5% suspension; positive control was given levamisole suspension 25 mg/kg BW and test group was given ethanol extract of African Leaves dose 100, 200, and 400 mg/kg BW. Mice previously induced by cow red blood cells 1% one day before given by treatments. Then given by treatments once day every day for 7 days. Data were analyzed by using SPSS version 17. The results showed that ethanol extract of African Leaves at dose 100, 200, and 400 mg/kg BW increased the hypersensitivity response and antibody titer of the male mice and showed significantly different with vehicle control CMC Na 0.5% (p < 0.5%). Dose 100 and 200 mg/kg BB not showed significantly different with positive control. Ethanol extract of African Leaves at dose 400 mg/kg BW showed better result as compared to dose 100 mg/kg BW and dose 200 mg/kg BW. Based on the results, it can be concluded that ethanol extract of African Leaves has the immunostimulatory effect.
- Imunomodulator merupakan substansi ataupun obat yang dapat memodulasi fungsi dan aktivitas sistem imun baik dengan cara merangsang ataupun memperbaiki fungsinya. Imunomodulator dapat diperoleh dari tanaman, salah satunya adalah Daun Afrika (Vernonia amygdalina Delile) yang mengandung flavonoid, glikosida, dan saponin yang dapat meningkatkan sistem imun. Tujuan penelitian ini adalah untuk mengetahui efek imunomodulator ekstrak etanol Daun Afrika terhadap respon hipersensitivitas dan titer antibodi sel imun mencit jantan. Penelitian ini dilakukan secara eksperimental dengan menguji respon hipersensitivitas dan titer antibodi sel imun mencit jantan. Uji respon hipersensitivitas dilakukan dengan mengukur volume pembengkakan telapak kaki mencit, sedangkan uji titer antibodi dilakukan dengan metode hemaglutinasi. Sebanyak 25 ekor mencit dibagi menjadi 5 kelompok yaitu kelompok kontrol pembawa diberi suspensi CMC Na 0,5%; kontrol positif diberi suspensi levamisol dosis 25 mg/kg BB dan kelompok uji diberi suspensi ekstrak etanol Daun Afrika dosis 100, 200, dan 400 mg/kg BB. Mencit diinduksi terlebih dahulu dengan sel darah merah sapi 1% sehari sebelum diberikan perlakuan. Kemudian diberikan perlakuan sehari sekali setiap hari selama 7 hari. Data dianalisis menggunakan program SPSS versi 17. Hasil penelitian menunjukkan bahwa ekstrak etanol Daun Afrika dosis 100, 200, dan 400 mg/kg BB dapat meningkatkan respon hipersensitivitas dan titer antibodi sel imun mencit dan menunjukkan perbedaan yang signifikan dengan kontrol pembawa CMC Na 0,5% (p < 0,05). Dosis 100 dan 200 mg/kg BB tidak menunjukkan perbedaan yang signifikan dengan kontrol positif. Pemberian ekstrak etanol Daun Afrika dosis 400 mg/kg BB menunjukkan hasil yang lebih baik dibanding dengan dosis 100 mg/kg BB dan dosis 200 mg/kg BB. Berdasarkan hasil di atas, dapat disimpulkan bahwa ekstrak etanol Daun Afrika mempunyai efek imunostimulator.
