- Soursop leaf (Annona muricata L.) is one of the Annonacae family used to treat various types of diseases. Soursop leaves contain acetogenin, tannins, alkaloids, flavonoids, and steroids. The aim of this study was to determine the antioxidant activity of ethanolic extract of young and old leaves of soursop with DPPH (1,1-diphenyl-2-picrylhydrazil) radical scavenging method. Young and old leaves of soursop were collected, cleaned, dried, and powdered, characterized and screened, then each powdered soursop leaf was extracted with maceration using ethanol 96%. The ethanolic extracts of young and old leaves of soursop were tested the antioxidant activity against DPPH as free radicals, DPPH absorbance was measured using visible spectrophotometer at a wavelength of 516 nm in the 10th minute after the addition of methanol solvent. The ability of antioxidants was measured as a decrease in absorbance of the DPPH solution after the addition of the extract. The results of powder characterization of young and old leaves of soursop obtained water content of 7,98%; 7,97%, levels of water-soluble extract 26,20%; 20,41%, levels of ethanol-soluble extract 20,25%; 14,89%, total ash content of 5,52%; 5,33%, and the ash content is not soluble in acid 1,32%; 1,14%. Phytochemical screening results of young and old soursop leaves simplisia contain alkaloids, flavonoids, glycosides, tannins, saponins and steroids. The results of antioxidant activity showed that ethanolic extract of young and old soursop leaves had half Inhibitory Concentration (IC50) of 46,77 ± 0,1156 μg/ml dan 44,37 ± 0,1350 μg/ml, respectively. While, vitamin C had IC50 of 4,24 ± 0,0115 μg/ml. Both extracts had lower antioxidant activity compared to vitamin C. The result showed that ethanolic extract of young and old leaves of soursop had antioxidant activity with very strong category.
- Daun sirsak (Annona muricata L.) merupakan salah satu tanaman dari famila Annonaceae yang digunakan untuk mengobati berbagai jenis penyakit. Daun sirsak mengandung acetogenin, tanin, alkaloida, flavonoida dan steroida. ekstrak etanol daun muda dan daun tua sirsak dengan metode pemerangkapan DPPH (1,1-diphenyl-2-picrylhidrazyl). Daun muda dan daun tua sirsak dikumpulkan, dicuci, dikeringkan lalu dihaluskan selanjutnya masing-masing serbuk simplisia dikarakterisasi dan diskrining, dan diekstraksi secara maserasi menggunakan pelarut etanol 96%. Ekstrak etanol daun muda dan daun tua sirsak di uji aktivitas antioksidan terhadap DPPH sebagai radikal bebas, absorbansi DPPH diukur menggunakan spektrofotometer sinar tampak pada panjang gelombang 516 nm pada menit ke-10 setelah penambahan pelarut metanol. Kemampuan antioksidan diukur sebagai penurunan absorbansi larutan DPPH akibat adanya penambahan ekstrak. Hasil karakterisasi serbuk simplisia daun muda dan daun tua sirsak diperoleh kadar air 7,98%; 7,97%, kadar sari yang larut dalam air 26,20%; 20,41%, kadar sari yang larut dalam etanol 20,25%; 14,89%, kadar abu total 5,52%; 5,33%, dan kadar abu yang tidak larut dalam asam 1,32%; 1,14%. Hasil skrining fitokimia daun muda dan daun tua sirsak mengandung senyawa kimia golongan alkaloid, flavonoida, glikosida, tanin, saponin dan steroida. Hasil pengujian aktivitas antioksidan menunjukkan bahwa ekstrak etanol daun muda dan daun tua sirsak memiliki nilai Inhibitory Concentration 50% (IC50) sebesar 46,77 ± 0,1156 μg/ml dan 44,37 ± 0,1350 μg/ml. Sedangkan, vitamin C memiliki nilai IC50 sebesar 4,24 ± 0,0115 μg/ml. Kedua ekstrak etanol ini memiliki aktivitas antioksidan lebih rendah dibandingkan dengan vitamin C. Hasil penelitian menunjukkan bahwa ekstrak etanol daun muda dan daun tua sirsak memiliki antioksidan dalam kategori sangat kuat.
