  • SUMMARY YULIA MAIDARNIS. The Effect of Using Accessions, Cryoprotectan Modifications and Type of Explants to Water Yam (Dioscorea alata L.) Explants Recovery with Vitrification Technique by In Vitro. (Mentored by Moh. Ana Syabana Amin, Susiyanti and Ragapadmi Purnamaningsih). The aim of this research was to know the effect of using accessions, cryoprotectan modifications and type of explants to water yam (Dioscorea alata L.) explants recovery with vitrification technique by in vitro. This research was done at Laboratory of Tissue Culture Cell and Tissue Biology Researchers Group (BSJ) BBBIOGEN, Bogor from June to August 2013. This research used three treatment factors. The first factor was water yam accessions with two levels, they were 748 accession (A1) and PDL 4 accession (A2). The second factor was the cryoprotectan modifications with four levels, they were LS 20 minutes PVS2 30 minutes (K1), LS 20 minutes PVS2 45 minutes (K2), LS 30 minutes PVS2 30 minutes (K3), and LS 30 minutes PVS2 45 minutes (K4). The third factor was type of explants with two levels, they were a knot of peak explants (E1) and two knots of stem explants (E2). From those factors there were 16 treatment combinations and repetad for 3 times so there were 48 treatment units in total. This research used descriptive analyzed and was shown in tabulation data. The result of this research showed that PDL 4 accession, LS 20 minutes PVS2 30 minutes cryoprotectan modification and two knots of stem explants gave the best living percentage, by in it self or combinations.
