  • Stevioside is a glycoside compound contained in stevia leaves which has a sweetness level 300 times sweeter than sugar and stevioside is non carcinogenic. Stevioside has no calories and less glycem index, makes it suitable as a natural sweetener for diabetic patients. Stevia mass production conventionally by seed and cutting stem, but it is not effective because the ability of seed stevia to germinate very low and plants earned from multiplication of cuttings is limited. This research used randomized design with two factorials and three replications. The combination treatments were level of 2,4-D : 0,5, 1, 1,5 mg/l and level of kinetin : 0, 0,5, 1, 1,5 and 2 mg/l. The result showed that the callus formed was reported in combination treatment 2,4-D 1 mg/l and kinetin 2 mg/l after 11 days culture. The best callus diameter was found 1,17 cm in combination treatment 2,4- D 1,5 mg/l and kinetin 1 mg/l after 6 weeks culture. The colors of callus were brown, white brownish, green brownish and green yellowish. The textures of callus were compact, intermediate and friable.
