- The research was aimed to know the effect of IBA concentrations and various explants to patchouli (Pogostemon cablin Benth) in vitro synthetic seed germination. The research was conducted in Biotechnology Laboratory Faculty of Agriculture of Sultan Ageng Tirtayasa University from October until December 2016. The research used Randomized Completely Design (RCD) which was arranged as factorial with two factors. The first factor was concentrations of IBA (A) with three levels : IBA 0 mg/l (A1), IBA 2 mg/l (A2), and IBA 3 mg/l (A3). The second factor was various explants with three levels : axillary buds (B1), nodal segments (B2), and internode (B3). There were 9 combination treatments and replicated 3 times. The result showed that concentration of IBA 2 mg/l gave the best result to patchouli synthetic seed germination in parameter of plant root’s length 4 week after planting (0.51 cm), plant’s height 4 week after planting (0.51 cm), number of sprout 2 week after planting (4,00 pieces), number of sprout 3 week after planting (6.00 pieces), number of sprout 4 week after planting (6.20 pieces), number of bud 2 week after planting (4,00 pieces), number of bud 3 week after planting (6.00 pieces), number of bud 4 week after planting (6.20 pieces). The used of axillary buds as explant showed the best result in parameter of plant root’s length (0.51 cm), number of sprout 2 week after planting (3.99 pieces), number of sprout 3 week after planting (6.11 pieces), number of sprout 4 week after planting (6.44 pieces), number of bud 2 week after planting (3.99 pieces), number of bud 3 week after planting (6.11 pieces), number of bud 4 week after planting (6.44 pieces). There was interaction between IBA concentrations and various explants to all parameters observed, except to parameters number of sprout 1 week after planting and number of bud 1 week after planting.
