  • Stevia (Stevia rebaudiana Bertoni) is a bushes plant that contains active compound of diterpene glycoside with level of sweetness between 200-300 times than sugar cane sucrose and do not contain calories so it is safe to be consumed by diabetics and for those who is in diet programme. Elicitor is a signal molecules that stimulate secondary metabolites form in cell culture. One type of biotic elicitor is amino acid. Amino acids have an important role for callus growth and differentiation. The purpose of this research was aimed to know the effect of type and concentration of elicitor combinations for Stevia rebaudiana callus induction in in vitro. This research used Completely Randomized Design (CDR) with one factor that consists of 12 levels X1 (caseine hydrolysate 100 mg/L), X2 (caseine hydrolysate 200 mg/L), X3 (caseine hydrolysate 300 mg/L), X4 (yeast extract 100 mg/L), X5 (yeast extract 200 mg/L), X6 (yeast extract 300 mg/L), X7 (glutamine 100 mg/L), X8 (glutamine 200 mg/L), X9 (glutamine 300 mg/L), X10 (proline 100 mg/L), X11 (proline 200 mg/L), X12 (proline 300 mg/L). The results showed that the combination treatment of type and concentration of elicitor gave real effect. The best combinations was concentration of glutamine 100 mg/L (X7) with the callus induction time 12 days and callus diameter around 1,08 cm in 6 weeks. Every callus in each treatment showed yellowish green in color with solid texture.
