- Nilam (Pogostemon cablin Benth.), is one of the aromatic plants that produce essential oil (Patchouli oil). This research aims to know the influence of growing regulatory substance auxin (2,4-D) and citokinin (BAP) against the formation of the callus and knowing the optimum concentration of BAP for the initiation of shoots of Nilam (Pogostemon cablin Benth.) varieties Patchoulina 2 via in vitro. This research has conducted in November 2016 until April 2017. This research was conducted in the agricultural seed development unit laboratory, Bogor. The experiment was conducted in two stages: callus induction and initiation of shoots. The design used in this research was completely randomized design (CRD) non factorial for initiation of shoots. The first factor in treatments for callus induction with concentration of 2,4-D were D0 (MS+0 mg/l 2,4-D), D1 (MS+ 1 mg/l 2,4- D), D2 (MS+2 mg/l 2,4D) and D3 (MS+3 mg/l 2,4-D). The second factor with concentration of BAP were B0 (0,1 mg/l BAP) and B1 ( 0,5 mg/l BAP). The treatments for initiation of shoots with concentration of BAP were), B1 (0,1 mg/l BAP), B2 (0,5 mg/l BAP), B3 (1,0 mg/l BAP), and B4 (MS+1,5 mg/l BAP) and B5 ( MS+2,0 mg/l BAP). The results showed medium D1B2 (MS+ 1 mg/l 2,4-D + 0,5 mg/l BAP) gives best results on time variables callus initiation (12 HST), white callus, crumbs callus and diameter of the callus ( 1,63 cm). At the intiation of shoots, medium MS+ 1,5 mg/l BAP was able to intiate shoots at 56 HST with shoots growth percentage 93,3 %.
