- Exploration Gene Resistance towards Bacterial Leaf Blight (Xanthomonas Oryza pv. Oryzae) on Rice Lines Based on Molecular Analysis and Resistance Test to Xoo Bacterial Isolates. Bacterial Leaf Blight (BLB) disease is important for rice cultivation, which causes high yield losses, particularly in the rainy season reaches 20.6 to 35.6%, while in the dry season can reach 7.5 to 23.8%. An effective way to overcome the disease is to develop a resistant varieties. This study aimed to identify the BLB resistance gene in rice lines by molecular analysis and to identify the level of resistance of rice lines to Indonesian Xoo bacterial leaf blight isolates.This research was conducted at the Laboratory of Molecular Biology and Greenhouse Indonesian Center Agricultural Biotechnology and Genetic Resources Research and Development, from September 2016 to February 2017. Material used were collection of rice derived from BB Rice of 44 numbers consisting of 12 varieties or selected rice lines, 14 IRRI isogenic lines, and 18 numbers of rice lines from crossing. The result showed that the rice varieties not only have a single gene but many genes (multigenic). The molecular marker-assisted selection process was divided into two stages. Initial stage by using the parent of isogenic and crossing (DQ1 and DQ2) and was continued by identification of rice strains from crossing result. Initial selection was aimed to identify Xa/xa resistance genes in varieties and parent, were compared to band patterns of PCR result on allele of resistance genes of IRRI isogenic lines, whereas further selection aimed to identify lines from crossing, were suspected having Xa/xa gene introgration from the crossing material, based on initial selection results. The resistance test was performed by using clipping method with Xoo12-183 isolate. The result showed that the parent DQ1 has the genes of Xa7 and Xa21, whereas the parent DQ2 has the genes of Xa2 and Xa21. The resistance reaction showed that the reaction was slightly susceptible to DQ1 and DQ2.
