- The objective of the research was determine of patchouli plants germination by the treatment of alginate and NAA(α-Naphthaleneacetic acid) concentration levels in vitro. The research was conducted in Biotechnology Laboratory of Faculty of Agriculture, University of Sultan Ageng Titayasa from March to May 2017. The research used a factorial of completely randomized design with two factors. The first factor was the level of alginate concentrations (A), there were: alginate 2% (A1), 3% (A2) and 4% (A3). Then, the second factor was the level of NAA (α-Naphthaleneacetic acid) hormone concentrations (N), there were: NAA 1 mg/l (N1), 2 mg/l (N2), 3 mg/l (N3). With 3 replications, 27 test units were obtained. Explant used was patchouli buds from tissue culture. Parameters observed were germination times, number of seed germinated, number of buds, number of roots, roots percentage, plant heigh and percentage of seed germination. The research showed that 4% alginate gave the best concentration on the parameters of number of germinated seeds (2,44 pieces), the number of shoots (2,67 pieces) and the percentage of germinated seeds (12,7%). The results showed that there was no interaction between given alginate and NAA concentration levels to all parameters.
