- Kemiri sunan (Reutealis trisperma (Blanco) Airy Shaw) is a kind of plant that can be the source of biodiesel because it can produce fruits and seeds that contain oil up to 50%. Through tissue culture techniques with methods of direct organogenesis and somatic embryogenesis in mean produced much plant faster. The purpose of this research was to know the effect of formulation medium on bud growth and callus growth of kemiri sunan in vitro. The design used in this research is completely randomized design (CRD). The treatments for initiation of bud from apical bud were (d1)1⁄2 WPM + Kin 0,1 mg/l, (d2) WPM, (d3) 1⁄2 WPM, (d4) WPM + BA 0,01 mg/l, (d5) WPM + BA 0,1 mg/l, (d6) WPM + 2-iP 0,01 mg/l, (d7) WPM + 2-iP 0,1 mg/l, (d8) 1⁄2 WPM + BA 0,01 mg/l, (d9) 1⁄2 WPM + BA 0,1 mg/l, (d10)WPM + 2-iP 0,01 mg/l, (d11)1⁄2 WPM + 2-iP 0,1 mg/l. The treatments for induction of embryonic callus from embryo were (m1) MS + 2,4-D 3 mg/l + Casein Hydrolysate 3 g/l, (m2)MS + ABA 0 mg/l + BA 0 mg/l, (m3) MS + ABA 0 mg/l + BA 1 mg/l, (m4) MS + ABA 0,5 mg/l + BA 0 mg/l, (m5) MS + ABA 0,5 mg/l + BA 1 mg/l, (m6) MS + ABA 1 mg/l + BA 0 mg/l, (m7) MS + ABA 1 mg/l + BA 1 mg/l. The treatments for formation of embryo somatic structure were (k1) medium induction of embryonic callus, (k2) WPM + BAP 0,5 mg/l + Kinetin 1 mg/l + glutamine 100 mg/l, (k3) WPM + BAP 0,5 mg/l + Kinetin 2 mg/l + glutamine 100 mg/l, (k4) WPM + BAP 0,5 mg/l + Kinetin 3 mg/l + glutamine 100 mg/l. The result showed that 1⁄2 WPM medium with Kinetin 0,1 mg/l is the best treatment in the leaf number (1,5 sheet) and medium 1⁄2 WPM + BAP 0,01 mg/l is the best treatment in the long of bud (1,10 cm) at initiation of bud from apical bud in. MS medium with ABA 0,5 mg/l + BAP 0 mg/l is the best treatment in callus weight (7,477 g) at induction of embryonic callus from embryo. WPM medium BAP 0,5 mg/l + Kinetin 1 mg/l + glutamine 100 mg/l is the best treatment in torpedo embryo number and increase of kinetin concentration can decrease the crumbs callus number at formation of embryo somatic structure.
