The aim of this research was to determine the response of Banten local rice accessions (Oryza sativa L.) to morphological and molecular test of salinity tolerance. This research was conducted from December 2018 to February 2019 at the Laboratory of Biotechnology, Agricultural Faculty of Sultan Ageng Tirtayasa University, Serang-Banten. The morphological test used a Randomized Block Design (RBD) with two factors. The first factor was the rice accessions consisting of five levels: A1 (Kewal Bulu Hideung), A2 (Pare Cerai), A3 (Tunggul Hideung), A4 (Inpari 35) and A5 (IR64). The second factor was salinity concentrations consisting of five levels: A1 (0 pp m), A2 (2000 ppm), A3 (4000 ppm), A4 (6000 ppm) and A5 (8000 ppm). Each treatment was repeated three times, to obtain the results of seventy-five experimental units. The parameters observed in this research were seedling plant height, wet weight, dry weight, germination, root length, maximum growth potential, and dendograms. The phases of molecular test started from DNA isolation, quantitative and qualitative DNA test, PCR amplification with three STS primers: Wn11463, Wn13900 and Th32639, agarose gel electrophoresis for PCR results, DNA bands scoring, and making dendograms. The result of the morphological test showed that the rice accessions gave different results on each parameters and there was an interaction between rice accessions and salinity concentrations to the parameters of germination (Cerai 88,89%) and maximum growth potential (Cerai 3%). The result of molecular test showed that the Kewal Bulu hideung and Cerai rice accessions had the character of salinity tolerance with the value of genetic similarity coefficient to inpari 35 was 0,64.
