The research was aimed to know the transformation of folate gene by the in planta method on rice (Oryza sativa L.) Banten local variety Tunggul Hideung. This research was conducted at the Biotechnology Laboratory of the Faculty of Agriculture, Sultan Ageng Tirtayasa University from February to April 2019. This study used a completely randomized design (CRD) consisted of 1 factor with 5 replications. The single factor used was hygromycin concentration consisted of 4 levels namely 25, 50, 75 and 100 mg/L so that there were 20 experimental units. Each experimental unit consisted of 5 explants so that 100 explants were needed in the study. Data were analyzed by analysis of variance and if the results showed significantly different, then further tests were carried out with Duncan Multiple Range Test (DMRT) at the level of 5%. From the results it was found that the treatment of hygromycin concentration was able to gave a significantly different effect on the transformation efficiency parameters and the percentage of explants that died in transformation in planta. The treatment of hygromycin concentration 25 mg/L is the optimum concentration by giving results that are significantly different from the parameters of the observed transformation efficiency of 64% and the percentage of explants to die by 36% in transformation in planta.
