This study was aimed to determine the effect of Benzylaminopurine and Thiamine on in vitro shoots multiplication of Japanese taro (Colocasia esculenta (L.) var. Antiquorum). This research was conducted from August to November 2018 at Plant Tissue Culture Laboratory of Center for Agriculture Production Technology – Agency for The Assessment and Application of Technology – Research Center for Science and Technology, South Tangerang - Banten. This research used a Completely Randomized Design with two factor and three replicant. The first factor was BAP concentration consist of four levels i.e. 0, 1, 2, and 3 mg/L. The second factor was Thiamine concentration consist of four levels i.e. 0, 1, 2, and 3 mg/L. Observation was done every once a week for 12 weeks. The results showed that the treatment of single BAP 2 mg/L was able to produce the highest number of leaves (5.29 leaves) and the number of shoots (5.33 shoots) and was the best concentration for shoots multiplication. Thiamine treatment 3 mg/L gave the best results on the parameters of the plant height (4.13 cm), number of leaves (2.92 leaves) and number of roots (9.12 roots). There is an interaction between BAP and Thiamine treatment in 10 Weeks After Planting on the number of shoots with a combination of BAP 1 mg/L and Thiamine 0 mg/L treatment with the highest value (3.28 shoots) and the combination of tuber diameter BAP 0 mg/L and Thiamine 2 mg/L treatment with the highest value (0.89 cm).
