  • Pogostemon cablin Benth. is plant which had economic value. So it was necessary to develop a method in order to accelerate multiplication of planlet. This research aimed to determine the effect of sucrose concentration and wavelength on growth and chlorophyll content of patchouli plant (Pogostemon cablin Benth.). The research was conducted from April until June 2015 in the Biotechnology Laboratorium of Faculty of Agriculture, Sultan Ageng Tirtayasa University, Serang, Banten. The culture medium was modified Murashige and Skoog medium with the addition of hormone BAP (0,5 mg/l). The research used Randomized Block Design (RBD) that consisted of two factors. The first factor of sucrose concentration consisted of four levels: S0 (0 g/l); S1 (20 g/l); S2 (30 g/l); S3 (40 g/l). The second factor consisted of five levels of illumination color: B0 (control), B1 (red), B2 (yellow), B3 (green), and B4 (blue). Therefore, there were 20 combinations of treatments, each treatment was repeated three times so there were 60 experimental units. The result showed that the addition of sucrose 30 g/l gave the best result on plant height, number of shoots, and number of leaves. Whereas the red convex plastic covers increased number of shoots, and plant height. Wavelength which provided the best growth nilam was red and blue. Concentration sucrose and wavelength didn’t give effect on chlorophyll content ratio.
