- This study aimed to develop Two-Tier Multiple Choice (TTMC) to assess higher order thinking skills on virus and qualify the validity, reliability, difficulty index, discrimination index, and distractor effectiveness. The research method used was research and development (R&D) and the design based on Borg and Gall (research and information collecting, planning, develop preliminary from of product, preliminary field testing I and main product revision, and preliminary field testing II). Data collection techniques used were interview data from teacher and students, written tests, and questionnaires. The results analysis were as follows: (1) TTMC has very good criteria, score an average of 85.08%; (2) The validity of 67% feasible category and 33% not feasible category; (3) reliability 0.85 was considered very high (4) the difficulty index 80% feasible category and 20% not feasible category; (5) the discrimination index 67% feasible category and 33% not feasible category; (6) the distractor effectiveness 10% very good, 13.3% good, 43.3% relatively poor, 16,7% poor, and 16.7% very poor. The student’s response to the TTMC showed 72.44% so that it can be categorized as good. Based on the results of the research, 9 of 15 items TTMC developed (1, 6, 8, 9, 10, 11, 12, 13, dan 14) can be used as an assessment instrument on virus material for students of class X.
