- Platinum group of chemotherapy drugs, especially oxaliplatin have several side effect. Chemotherapy drug can induce oxidative stress in biological system. Increase of ROS can cause mitochondrial disfunction which causes microglia to release proinflammatory mediators which causes cell apoptosis. The apoptosis marker can be characterized by caspase-3 protein. Therefore, this study aimed to observe the effect of quercetin at the expression of caspase-3 in CIPN mice induced by oxaliplatin in the thalamus and S1 area. In this study 9 mice were grouped in 3 groups consisting of a control group, CIPN group and CIPN with quercetin 100 mg/kgBW group. Dosage of oxaliplatin to induce a CIPN model was 3 mg/kg administered on day 0, 2, 4 and 6, while quercetin administered per day for 7 days in treatment group. Mice were sacrificed on day 22, brain were extracted, somatosensory cortex (S1) and thalamus area were analysed. After obtaining the required brain area, total RNA was extracted using a RNA mini kit, then continued by converting mRNA to cDNA and amplification by polymerase chain reaction (PCR). Caspase-3 expessions in CIPN group increased higher than in control group, both in thalamus and S1 (p > 0,05). While in treatment with quercetin group, caspae-3 expression tend to decrease than CIPN group, both in thalamus and S1 (p > 0,05). It can be concluded that quercetin 100 mg/kgBW in CIPN mice induced by oxaliplatin tend to make caspase-3 expression decreasing in thalamus and S1.
