  • Objective : to evaluate limbal mesenchymal stem cell conditioned medium (LMSC-CM) on cell density and caspase-3 expression of corneal endothelial cells after exposure to phacoemulsification ultrasound energy in rabbit. Material and methods: Twenty-four New Zealand white rabbits (24 eyes) were divided randomly into 2 groups. The control group was treated with intracameral balanced salt solution (BSS) after exposed to phaco ultrasound, while the treatment group was treated with intracameral LMSC-CM. Endothelial cell density were evaluated with specular microscopy before ultrasound exposure and on 3rd day thereafter. Cornea were excised on 3rd day to evaluate expression of caspase-3 in endothelial cell by immunohistochemistry staining. Results: corneal endothelial cell density changes were -93.69 ± 139.04 in the control group and -119.31 ± 85.41 in the treatment group (p = 0.04 and p = 0.001, respectively) showing that ultrasound exposure significantly reduced corneal endothelial cell density in both groups. While the comparison of changes between groups did not differ significantly (p = 0.592). Likewise, endothelial caspase-3 expression in the treatment group was also not significantly different compared to the control group on the 3rd day histopathology examination, with a mean of 2.17 ± 0.777 and 1.17 ± 0.322, respectively (p = 0.625). Conclusion: LMSC-CM administration did not affect endothelial cell density and caspase-3 endothelial expression of corneal rabbits after exposure to ultrasound phacoemulsification.
