- This study aims to determine the molecular weight of S protein (Spike Glycoprotein) isolates of Massachusset and field isolate I-147/11 using SDSPAGE and Dot Blot. Whole virus in the form of allantois liquid was performed electroforesis and then the results were calculated by standard protein regression formula to find the molecular weight of S protein, then the result continued with Dot Blot test using whole virus and protein virus to ensure protein S purify and using molecular weight of fertile allantois liquid based on previous research. The results of this study indicate the weight of S protein Massachusset and field isolate I-147/11 were 168.21 kDa, so there is no difference molecular weight of the virus. Protein S (Spike Glycoprotein) IBV isolate Massachussets and local isolate I- 147/11 can be identified with Dot Blot.
