- An experimental study to determine histopathological changes of aorta in cirrhosis rats following treatment with the endotoxin of Escherichia coli (E. coli) O 55:B was carried out. A completely randomized design with 5 treatments (each of which replicated 5 times) was used in this study. In control treatment (PO), cirrhosis rats were treated only with 0.9% NaCl solution and the histopathological changes of aorta were observed 6 hours following treatment. In endotoxin treated groups, cirrhosis rats were injected with the endotoxin at the doses of 3 mg/kg body weight and the histological changes of aorta were respectively observed at 6 (PI), 12 (P2), 18 (P3), and 24 (P4) post injection. The parameters observed were the number of endothelial cells per cm aorta, the rate of discontinuity in the internal elastic lamina, and the thickness of the internal elastic lamina. The result showed that the number of endothelial cells were significantly (P<0.05) lower in endotoxin treated rats than in the controls, i.e. PO (6.6920±0.1205 cells/cm), PI (6.2640±0.2281 cells/cm), P2 (6.2900±0.4600 cells/cm), P3 (2.980±1.4777 cells/cm), and P4 (1.0020±0.3932 cells/cm). The discontinuity of internal elastic lamina was significantly (P<0.05) higher in endotoxin treated rats than in control groups, i.e. 0,00 in PO, PI, and P2, 0.667±0.147 um in P3 and 2.312 ±0.892 urn in P4. A significant reduction in the thickness of internal elastic lamina was also observed in rats treated with the endotoxin, i.e. PO (0.381±0.047 ,zrn), PI (0.397±0.062 um), P2 (0.365±0.048 gm), P3 (0.174±0.063 um), and P4 (0.122±0.029 pm). It is evident that the endotoxin of E. coli 055 : B 5 can cause a significant damage to the aorta of cirrhosis, especially in prolonged exposure of the endotoxin.
