  • Introduction. The usage of cigarette has been a huge problem towards the medical field worldwide. Cigarettes are carcinogens, thus smoking problems has a wide impact to the smoker and also the environment surrounding the smoker who become passive smokers. Oral squamous cell carcinomas start from a dyplasia that is not prominent clinically, thus a biomarker is highly needed to detect the dysplasia condition before turning into cancer. Matrix Metalloproteinase (MMP-9) is a cancer marker that is often studied but a study expression of MMP-9 on dysplasia condition due to cigarette smoke exposure not yet been proved. Purpose: To prove the increase of MMP-9 expression towards the mucosa affected by displasia because of cigarette smoke exposure as a risk factor of oral cancer. Method: The study used wistar mice (Rattus novergicus) that was exposed by cigarette smoke for 4 weeks, then 8 weeks after dysplasia observation was performed using hematoxylin Eosin (HE) staining and the expression of MMP-9 observed by the immunohistochemistry staining which then compared with the control group. Result: Dysplasia occurs according to the length of cigarette smoke exposure and also the increase of MMP-9 expression towards the group with dysplasia. Conclusion: A strong correlations has been occured between the increase of MMP-9 expression towards the dysplasia level on the oral cavity epithel of the cigarette smoke-exposed wistar mice.
