- Acyclovir is a purine nucleoside analogue antiviral drug that has problem on its solubility and bioavailability. Cocrystal is a crystalline solid material modification method for changing the physical properties of the ingredients, especially the solubility properties of the drug. This study aimed to compare the methods Grinding and slurry on the characteristics of the formation of the acyclovir - succinic acid cocrystal by solvent evaporation method. On the formation of cocrystal acyclovir - auccinic acid Grinding method, based on the analysis of X-ray diffraction, infrared spectroscopic analysis, and the calculation of ED15 values known that grinding time 15 minutes was a time the most optimal grinding. In cocrystal Slurry method cocrystal, solvent concentration of 12 ml/g indicate the optimal concentration of solvent were added. The result of diffractogram profiles comparison between the methods of grinding and slurry with diffractogram profiles cocrystal solvent evaporation known that both the grinding and slurry method had a different profile to the diffractogram profile of solvent evaporation cocrystal. Comparisons of ED15 was known that there a significantly increased percent of acyclovir dissolved in solvent evaporation and slurry method cocrystal with the concentration of solvent of 12 ml/g as compared to pure acyclovir and physical mixture of acyclovir and succinic acid.
