- The aims of this study was to identify Aeromonas salmonicida as the cause of Furunculosis disease in gouramy (Osphronemus gouramy) using conventional and polymerase chain reaction technique. Ten gills and skin swab samples were obtained from suspected Furunculosis gouramy. The samples were isolated on Trypticase Soy Agar and MacConkey Agar, than the colonies from that medium followed by identification tests using Triple Sugar Iron Agar (TSIA), Sulfide Indole Motility (SIM), Simmons Citrate Agar (SCA), Urea Agar, and Sugar Fermentation Test. Sugar fermentation tests that were used in this study was consists of Glucose, Lactose, Sucrose, Maltose, and Manitol. From these test, 3 out of 10 samples were identified as Aeromonas salmonicida. Based on the biochemical tests result that the bacteria could be determined as Aeromonas salmonicida, the PCR test was done for the confirmation. Ferric siderophore receptor gene were used as specific gene to identify Aeromonas salmonicida. The confirmatory test shows band at 389 bp.
