  • Synthesis and Analgesic Activity Test of 5-methyl-2-(4’- methylbenzoyloxy)benzoic acid in Mice (Mus musculus) Vagen Setya Liani The purpose of this experiment are to obtain 5-methyl-2- (4’-methylbenzoyloxy)benzoic acid as analgesic from salicylic derivative. The compound was synthesized by reacting 5-methyl salicylic acid and 4-methylbenzoyl chloride in THF as solvent. This reaction yield was 54,6% product. The melting point of the compound was 126-128°C. The structure of the compound was analyzed by UV spectrophotometer, IR spectrophotometer, and NMR-1H spectrometer. According to the spectroscopy data, the synthesized compound was 5-methyl-2-(4’- methylbenzoyloxy) benzoic acid. The analgesic activity of the compound was tested by writhing methode. The results showed that ED50 of 5-methyl- 2-(4’-methylbenzoyloxy)benzoic acid was 81,67 ± 12,53 mg/kgBB while ED50 of aspirin was 87,87 ± 16,32 mg/kgBB, which was not significantly different each other. It was concluded that 5-methyl-2- (4’-methylbenzoyloxy)benzoic acid had analgesic activity same as aspirin.
