  • METHOD VALIDATION OF VITAMIN C AND FOLIC ACID SIMULTANEOUS ANALYSIS WITH HIGH-PERFORMANCE LIQUID CHROMATOGRAPHY Tiara Vista Ramadhani A high-performance liquid chromatography (HPLC) method has been developed for simultaneous analysis of vitamin C and folic acid. Optimum condition used a Li-Chrospher reversed-phase octadecylsilyl column (150 x 4,6 mm; particle size 5 μm) for the separation of vitamin C and folic acid. Buffer 50mM potassium dihydrogen phosphate pH 6,5 and methanol ( 90:10 ) used as the mobile phase with flow rate 1,5 ml min-1 and 281 nm as the detection wavelength. As the result, this method fulfilled the requirement of method validation (specificity, linearity, accuracy, and precision). The method showed good specificity and linearity with Rs value 15,325 ; r vitamin C = 0,9952 ; Vxo vitamin C = 4,13% ; r folic acid = 0,9999 ; Vxo folic acid = 0,5%. The recoveries of vitamin C and folic acid were in range of 99,6% - 101,4% with CV of vitamin C = 0,50% and CV of folic acid = 0,37%. Ionic strength, pH, and quantity of buffer in mobile phase become the critical point of simultaneous analysis of vitamin C and folic acid.
