  • A research on isolation of secondary metabolite from fungi symbiotic Haliclona fascigera (WR3) and the test of its activity against pathogenic bacteria, Staphylococcus aureus and Methicillin Resistant Staphylococcus aureus (MRSA) has been conducted in the Biota Sumatera Laboratory, Andalas University. That was identified with the molecular determination 18S rRNA gene using a PCR (Polymerase Chain Reaction). The result of identification showed that symbiotic fungi (WR3) was similar with Penicillium oxalicum. Fungi P. oxalicum was grown on Sabouraud Dextrose Agar media then cultivated in rice media for 30 days. The result of cultivation was extracted with ethyl acetate and extract was fractionated with methanol and hexane. Secondary metabolites contained in semipolar fraction was separated using column chromatography with sephadex LH-20 and silica gel 60 as the stationary phase. The mobile phase was eluted by isocaratic and step gradient polarity method, respectively. Based on the result of column chromatography, it was collected two pure compounds (A.1and A.2). The form of A.1 was a crystal (1124,5 mg) with Rf value 0,46 using ethyl acetat: dicloromethane (1:4). The form of A.2 was a needle yellow crystal (164,2 mg) with Rf value 0,38 using ethyl acetat: dicloromethane (1:4). A.1 and A.2 positive for phenolic test (FeCl3). Further characterization of A.1 and A.2 was conducted by spectroscopic method of UV-Vis, Infrared, HPLC and its activity against Staphylococcus aureus and MRSA using agar diffusion method. Both of compounds showed antibacterial activity against Staphylococcus aureus (SA) and MRSA, with Minimum Inhibitor Concentration (MIC) of A.1 and A.2 againts SA is 125 μg/disk and 500 μg/disk respectively and MIC of both compounds against MRSA is 250 μg/disk and 500 μg/disk, respectively.
