- A coumarin was isolated from the ethyl acetate extract of hamerang stem bark (Ficus fulva Reinwardt Ex Blume). The stem bark of hamerang is extracted successively by hexane, ethyl acetate and methanol which was had antibacterial activity to Escherichia coli and Staphylococcus aureus. Ethyl acetate extract was separated by column chromatography using silica gel as the stationary phase and hexane, ethyl acetate and methanol as the mobile phase by step gradient polarity (SGP) system. Isolated compound is 0.0101 g dark yellow color which soluble in methanol. It was identified by thin layer chromatography (TLC) shown a blue fluorescence spot at ultraviolet (UV) 365 nm and lighter by NaOH 1% additon. The purity of compound observed by single spot on TLC with various composition of eluent. The UV spectra shown maximum absorption at a wavelength of 280 nm indicated electron transition from π to π* and 318 nm indicated electron transition from n to π*. IR spectrum shown absorption at wave numbers 3340, 1639, 1549-1410, and 1021 cm-1 which indicated the –OH group,C=O group, benzene ring, and presence of the C(O)–O .
