  • Isolation of coumarin compound from extract of Glodokan stem (Polyalthia longifolia) has been conducted. Air-dried powdered of stem was extracted with n-hexane, ethyl acetat, and methanol by using maceration method. Extract of ethyl actat was subjected to siliga gel chromatography colomn by using n-hexane, ethyl acetat, and methanol to afford 17 fractions. Fraction 11th (fraction K) was recrystallized with ethyl acetat, n-hexane, and chloroform, and then the compound was isolated by using preparative thin layer chromatography method with n-hexane : ethyl acetat (4;6). White powder isolated compound was tested its purity with TLC by using NaOH 1%, the result showed blue flourescence. Based on the result of characterization, isolated compounds were obtained classified into coumarin compound of scopoletin group. Extract ethyl acetate was tested antioxidant activity. The test results IC50 value of 220,35 mg/L showed moderate antioxidant activity and Phenolic total value of 229,68 GAE/10mg dry extract.
