AKTIVITAS ANTIOKSIDAN SENYAWA METABOLIT SEKUNDER DAUN PALA (MYRISTICA FRAGRANS HOUTT)

  • ABSTRAKMetabolit sekunder daun pala diisolasi dengan maserasi 3500 g serbuk daun pala menggunakan pelarut metanol dan diperoleh ekstrak sebanyak 535,7 g (15,3%) dengannilai IC50 188,4 ppm, dilanjutkan partisi dengan pelarut n-heksana, etil asetat dankloroform, diperoleh ekstrak masing-masing 1,8488 g (0,35%), 0,501g (0,15%) dan7,122 g (2,28%) dengan nilai IC50 96,67 ppm, 0,216 ppm dan 35,62 ppm. Ekstrak nheksanadan kloroform diisolasi lebih lanjut dengan kromatografi kolom gravitasimenggunakan eluen masing-masing n-heksana : etil asetat (9:1) dan kloroform : etilasetat (8:2) dengan hasil diperoleh masing-masing 7 fraksi gabungan (MFHD 1-7) dan 6fraksi gabungan (MFKD 1-6) dimana seluruh fraksi gabungan aktif sebagai antioksidan.Fraksi gabungan dengan aktivitas tertinggi yaitu MFHD 2 (IC50 96,716 ppm), MFHD 4(IC50 0,729 ppm), MFKD 4 (IC50 69,132 ppm) dan MFKD 5 (IC50 direkromatografi dan dimurnikan dengan KLT preparatif, diperoleh fraksi dengan polanoda tunggal pada kromatogram KLT yaitu MFHD 2.3 (5,4 mg), MFHD 4.4 (10,7 mg)dan MFKD 4.4 (5,8 mg) dengan IC50 masing-masing 42,299 ppm, 44,084 ppm dan111,91 ppm. Hasil analisis GC-MS fraksi MFHD 2.3 mengandung senyawa eucalyptoldan sitostenone. Berdasarkan beberapa referensi kedua senyawa tersebut memilikiaktivitas antioksidan.Kata Kunci: Daun pala (Myristica fragrans Houtt), Aktivitas antioksidan 80,44 ppm) ABSTRACTIsolation of secondary metabolites nutmeg leaf maceration begins with 3500 g nutmeg leaf powder using methanol and obtained extract as much as 535.7 g (15.3%) with IC50value of 188.4 ppm, followed partition with n-hexane, ethyl acetate and chloroform,each extract was obtained 1.8488 g (0.35%), 0,501g (0.15%) and 7.122 g (2.28%) withIC50 value of 96.67 ppm, 0.216 ppm and 35.62 ppm. The extract n-hexane andchloroform were isolated further by column chromatography gravity using eluent eachn-hexane : ethyl acetate (9 : 1) and chloroform : ethyl acetate (8 : 2) with the resultsobtained by each of the 7 fractions combined (MFHD 1-7) and 6 fractions combined(MFKD 1-6) in which all of the combined fractions are active as antioxidants. Thecombined fraction with the highest activity MFHD 2 (IC50 96.716 ppm), MFHD 4(IC50 0,729 ppm), MFKD 4 (IC50 69.132 ppm) and MFKD 5 (IC50 80.44 ppm) returncolumn chromatography gravity and purified by preparative TLC, the fraction with apattern single spot on TLC chromatograms are MFHD 2.3 (5.4 mg), MFHD 4.4 (10.7mg) and MFKD 4.4 (5.8 mg) with IC50 respectively 42.299 ppm, 44.084 ppm and111.91 ppm. The GC-MS analysis of MFHD 2.3 fraction contains eucalyptol andsitostenone compounds. Based on some references both of these compounds haveantioxidant activity.
