  • Kapehu (Guioa diplopetala) are often used as traditional medicine at East Sumba. However, has not been much research about kapehu so that phytochemical compounds of kapehu is unknown. This study aims to screen potentially bioactive, analyze the antioxidant and antibacterial activity of kapehu, and the relationship between phenol, tannin and flavonoids content with their antioxidant and antibacterial activity of kapehu. The sample used in the form of 96% ethanol extract and methanol extract of the kapehu. The extract obtained by maceration method. The result of phytochemical screening known that kapehu contained some bioactive agents such as alkaloid, flavonoid, terpenoid/steroid, tannin, saponin and antrakuinon. Analysis antioxidant activity with DPPH (1,1-diphenyl-2-picrylhidrazil) method showed that methanol extract had higher value of antioxidant than ethanol extract. Antioxidant capacity of methanol extract was 82,3275 mg AEAC/g and ethanol extract was 59,58 mg AEAC/g. Antimicrobial activity with paper disk diffusion method showed that methanol extract from kapehu leaves extract has antibacterial activity on Escherichia coli and Staphylococcus aureus. It could be seen from the bacterial inhibition zone diameter of 6,1 mm for the bacteria Escherichia coli and 5,7 mm for bacteria Staphylococcus aureus. While, ethanol extract from kapehu leaves extract has not antibacterial activity. Phenol, flavonoid and tannin content have correlation with their antioxidant and antimicrobial activity.
