  • Background: One of stress’ impacts to health is the decrease of cellular immunity. BCG vaccination can enhance cellular immunity response. The aim of this study was to find out the role of BCG vaccination on cellular immunity on Balb/C mice against electric foot stress (EFS) and Listeria monocytogenes, examined from the number of spleen’s multinucleated giant cell (MGC). Methods: This study adapted laboratory experimental and Post Test Only Control Groups Design. Eighteen female BALB/c mice were divided into three groups and received standard laboratory diet. The first group (control/K) received no other additional treatment. The second group (Stress/S) received stress from EFS from day 12th until day 21st. The third group (stress-BCG/SB) received intra peritoneal injection of 0.1cc BCG at day 1st and 11th and stressed with EFS from day 12th until day 21st. At day 21st, all groups were injected intravenously with 104 live Listeria monocytogenes and sacrificed at day 26th. The spleen was taken. The MGC counting was done on 100 fields with microscope, 400 times magnification. The data were analyzed with One-Way ANOVA and Post Hoc Test Bonferroni. Result: There were significant differences on the number of MGC between K and S group (p=0,012), between K and SB group (p=0,000), and between S and SB (p=0,002). The lowest MGC number was in SB group while the highest was in K group. Conclusion: BCG vaccination significantly decreases the number of MGC in the spleen of Balb/C mice against EFS and Listeria monocytogenes. Key words: BCG, Stress, MGC
