  • Most of the Anophelines as malaria vector in the South and South-East Asian countries have been identified as cryptic species. Morfological difference of cryptic species is minute and may be restricted to a particular stage in the life-cycle. Therefore morphological identification might be misleading to characterize and identify cryptic species. Anopheles balabacensis is a member of the Leucosphyrus complex, Leucosphyrus Group. It has come to be recognized as important human malaria vector in Purworejo. Molecular studies using DNA barcode were carried out to investigates species recognition/identity and evolutionary relationship. DNA barcode that has been used to clarify closely allied species are ITS2 segment of ribosomal DNA and COI gene of mitochondrial DNA. In this study, we identify An.balabacensis from Purworejo morphologically and molecularly used ITS2 segment and COI gene. DNA barcode sequence of An.balabacensis were confirmed with GenBank database. Phylogenetic analysis was performed to know sequence character and to phylogenetic tree reconstruction. According to morphological identification, we found 279 sample of An.balabacensis from Purworejo. The adult female mosquito is periodically nocturnal (peak on 22.00-23.00), exophilic, antrophofilic, and most of them found on housing area near forest. There are 59,26% from 54 samples that molecularly as An.balabacensis. Based on gel electrophoresis, length of ITS2 segment is ±848 bp, further research is needed to make sure it. ITS2 segment of An.balabacensis are highly similar to An. dirus (87,16%), both of them are belong to Leucosphyrus Group. According to ± 700 bp of COI gene, the sample nearly identical (â�¤90%) with An.lutzii, An.albitarsis, An.guarani, or An.jeyporiensis. Intraspesific variation of ITS2 segment is 0%, whereas COI gene is 0,55%. Based on ITS2 segment, topology of phylogenetic tree beetween An.balabacensis and Leucosphyrus Group are monophyletic. Different result on phylogenetic tree topology based on COI gene, both of them are in separate clade or polyphyletic. Population of An. balabacensis from Purworejo showed genetic variation relatively less than speciation threshold point. Bionomics of populations An.balabacensis are not different each other. Nearest similarity of An.balabacensis is with members of Leucosphyrus Group.
