- Poly-Î2-hydroxybutyrate (PHB) is an organic polymer, which synthesized by many bacteria and serves as internal energy. PHB is potential as future bioplastic but its price is very expensive due to glucose usage in PHB industry. The development of PHB production using starch as an alternative carbon source has been conducted to reduce the dependence of glucose in PHB production. The aims of this research were: to isolate amylolytic bacteria which can produce PHB by using canna and arrowroot starches, to find out the optimum condition of selected isolate in producing PHB and the possibility of canna and arrowroot starch as an alternative carbon source in PHB production. This research was done in 5 steps: (i) amylolytic bacteria isolation, (ii) selection of the amylolytic bacteria based on hydrolytic ability, amylase activity and PHB producing capability, (iii) optimization of fermentation condition, (iv) polymer characterization and (v) identification of selected isolate. As many as 157 isolates were isolated and 56 among them were amylolytic. The amylolytic isolates were selected based on hydrolytic ability, amylase activity and PHB producing capability.
