- Durian is a native fruit from Indonesia that have highly economic value. Genetic variation and phenetic relationship between durian cultivar could be determine using Random Amplified Polymorphic DNA (RAPD) technique, easier and faster than other technique. The purpose of this reasearch were to identify and analyze genetic variation and phenetic relationship of durian cultivar in Java based on RAPD marker. The sample were durian leaf from 20 cuktivar that collected from 4 province in Java and as East Java, Central Java, Yogyakarta, and West Java. DNA was isolated from leaf using Nucleon-Phytopure, and was amplified by PCR using 7 random primers. The PCR product analyze using electrophoresis gel agarose 1,8% and visualized using UV iluminator. DNA banding pattern scored as binary matriks data and dendogram was constructed using Unweighted Pair Goup Method Arithmetic (UPGMA) (method from Numerical Taxonomy and Multivariate System (NTSYS) 2.1pc version. Based on DNA banding pattern, the polymorphism percentage was 83,69%. It showed that durian plant in Java have highly genetic variation. The dendogram showed 2 cluster with similarity coeficient 0,70-0,90. Durian from East Java is clustered into first cluster while durian from West Java clustered into second cluster, whereas durian from Central Java and Yogyakarta are separated into both cluster.
