  • Eucalyptus pellita F. Muell. is one type of eucalyptus that natural distribution in Indonesia. This plant belongs to the family of widely distributed Myrtaceae and it prospects for Industrial Plantation Forest (HTI). The plant has economic value that is used making materials pulp and paper. The aim of this research was to determine the concentration of ethanolic leaf extract of periwinkle that able to induce polyploydization and also analyze chromosomal characterization of Eucalyptus pellita F. Muell. The method steps used in the preparation of eucalyptus chromosome were squash, the steps fixation, 2N HCl to maceration and staining. The result showed that the ethanolic leaf extract of periwinkle (Catharanthus roseus (L.) G. Don.) at concentration of 1% is the optimal condition to induce Eucalyptus pellita F. Muell. become autotetraploid (2n=2x=44). The results of chromosomal characters can be determined by karyotype and idiogram. The result showed formula karyotype of Eucalyptus at concentration 0.1% and 0.5% were 2n=2x=22m, whereas the concentration of 0.05% is 2n=2x=18m +4sm. However, for concentration of 0.01% obtained formula of 2n=2x=20m+2sm. Karyotype in the control formula is 2n=2x=16m+6sm, whereas at concentration of 1% is 2n =4x= 40m +4sm.
