- Jembrana disease is an acute disease of Bali cattle (Bos javanicus) caused by Jembrana disease virus (JDV). The availability of early, sensitive and convenient detection methods will prevent disease outbreaks, economic losses to allowing routine controls of cattle health status. In this study, the possible use of subunit gag-ca gene as hybridization probe was evaluated. The gene was cloned in pGEX-CA and the JDV probe was synthesized from fragmen gag-ca gene using PCR labelled DIG to produce JCI a non-radioactive DNA probe, 208 bp. long. Based on BLAST analysis, the JCI probe has high degree of conservation amongst the Indonesian JDV isolates. In this study, the JCI probe is suitable for the detection of all the isolates. Dot-blot hybridization was carried out to detect the total RNA virus extracted from three cattles samples of spleen and two of blood samples of JDV-infected and one non-infected Bali cattle. The ten blood samples from Kalimantan of Bali cattles were disigned JDV infected. Results showed that the method was able to detect the JDV in infected animals originating from different areas. It was also able to detect the virus concentration as low as 10 pg as well. It can be concluded that gag-ca-based Dot-Blot hybridization technique is an efficient and specific diagnostic tool for Jembrana disease and it is suitable and applicable for conditions in which sophisticated equipments are not available.
