The technique of in vitro culture using a combination of anorganic fertilizer and organic complex casein hydrolisate as culture medium were develop for micropropagation. The aims of this research are to know whether anorganic fertilizer medium can be used as that of NP medium, to decide the concentration of casein hydrolisate should be added to anorganic fertilizer medium to improve the growth in vitro culture of Spathoglottis plicata Blume embryos. In this research, combination between (0, 1, 2, 3) gr/l anorganic fertilizer with a series (0, 25, 50, 75, 100) mg/l concentration of casein hydrolisate were used as in vitro culture medium. The experiments were repeated 3 times. Morphological characters were determined on the length of embryo(s) and number of leaves, also colour of developping embryo (protocorms). Quantitative data were analyzed using analysis of variance (ANAVA) and 1 % DMRT (Duncan&#xE2;&#xFFFD;&#xFFFD;s Multiple Range Test). The Result showed that 1 gr/l of Gw 32-10-10 + 75 mg/l casein hydrolisate and 1 gr/l of Gd 20-15-15 + 50 mg/l casein hydrolisate anorganic fertilizer can be used as culture medium of S. plicata orchid with 57.6 % and 61.5 % growth frequency, respectively.</description><publisher>[Yogyakarta] : Universitas Gadjah Mada</publisher><date>2011</date><type>Thesis:Thesis</type><type>PeerReview:NonPeerReviewed</type><identifier> , TRI SARTIKONINGSIH,S.P. and , Dr. Endang Semiarti, M.S.
