- Various media compositions have been formulated to optimize germination of orchid seed cultured and development of Spahtoglottis plicata plantlets. Modifications of fertilizer solution such, Growmore and Hyponex with various agar brand can be used on orchids seed culture. The objective of this research were to germinate the orchid seed in cultured and development of S. plicata plantlets by using the medium consist of fertilizer and agar brand, to determine optimum concentration of fertilizer and agar brand in germination of orchid seed cultured and development of S. plicata plantlets. The research was conducted using Completely Randomized Design (CRD). The first factors are 5 media type of inorganic fertilizer: Growmore 1 g/l (G1), Growmore 3 g/l (G3), Hyponex 1 g/l (H1), Hyponex 3 g/l (H3) and Vacin & Went (VW) as control, and the second factors are agar brands: Bioagar (B), Nutrijel (N) dan Swallow (S). Each treatment consisted of 3 replications. Data were analyzed with Analyze of Variance (Anova) a statistical test using Duncan Multiple Range Test (DMRT) at 5% level. The results showed that the fertilizer and agar brand shown the highest of germination percentage (97,64 and 96,17%) giving in medium H1S and VB:
