- The injured of bone is caused by several factors, such as trauma, tumors, and accident, thus it makes the continuity disruption of bone. In dealing with injured bone, it requires a specific approach to support regeneration process. One of it is by bone graft. Bone graft is a biomaterial alternative that can replace broken or missing bone. Hydroxyapatite (HA) of local material has been developed including an effort to refine its characteristics by composite manufacturing process, yet before the material used it needs an in vivo test by using experimental animals first. The aim of this research is to know the effect of CoHA-Gelatin-PVA and HA200-Gelatin-PVA composite as comparator implanted in femur bone of Wistar mice in speeding up the callus formation of bone. The research begins with implanting CoHA-Gelatin-PVA and HA200-Gelatin-PVA composite as comparator in femur bone of Wistar mice. The control group of femur bone is just hollowed out. Thirty white tailed Wistar mice are divided into six treatment group. The white Wistar mice are being euthanasia in the day of 14 and 21. The femur tissue is collected and prepared for histology observation to see bounded tissue, hyaline cartilage, woven bone and trabecular bone that fill callus by scoring method. Then, the specimen is being colored by using Malorry painting. The observed specimen is observed by using light microscope with 400x of zooming. The statistic analysis by Kruskal Wallis test to know the differentiation of treatment group, if there is a differentiation, so it is continued with Mann Whithey test. The result of statistic test shows that there is a meaningful differentiation (p<0,05) at the day of 14 and 21, where the treatment group has the greater callus number than it compares with control group. While there is a significant differentiation, the average result of callus number shows that the highest callus number on the group of 14 days is the treatment group of HA200- Gelatin-PVA composite and the lowest is control group. In the group of 21 days, the highest callus number is the treatment group of HA200-Gelatin-PVA composite and the lowest value group is control group. Based on the research result, it can be summed up that the giving of HA200-Gelatin-PVA composite implanted in white Wistar mice is more effected in speeding up of callus formation, where the callus number is greater than the giving of CoHA-Gelatin-PVA composite.
