- The product of tomato and maize transgenic is necessary to be distinguished from non-transgenic plants. Therefore, it is need to develope a method for identifying these tomato and maize transgenic. One of the methods that was developed in this work was, by identifying the35S CaMV promoter fragment using Polymerase Chain Reaction (PCR), since most transgenic plants use this type of promoter for expressing the gene interest. In PCR method, the purity of DNA fragment plays an important role to get the good result. Generally, DNA extraction is carried out by using kit method PureLink TM Plant Total DNA Purification Kit. The result of DNA extraction and purification indicated that, from tomato A, B and C there were obtained 0.11% (b/b), 0.12 % (b/b) and 0.17 % (b/b) of DNA with the purity level of 1.79, 1.75 and 1.74 respectively. In the DNA obtained of maize A, B and C were 0.41 % (b/b), 0.42 % (b/b) and 0.43 % (b/b) with the purity level of 1.76, 1.74 and 1.77 repectively. Visualization result of 35S CaMV amplification with the annealing temperature of 55°C and a number of template 300 ng demonstrated high intensity fragment for tomato B and maize B, whereas no amplification product for tomato A, tomato C, maize A and maize C. It could be stated that tomato B and maize B contained 35S CaMV promoter.
