Histological Study of Skin Aging Model Treated with Topical Mesenchymal Stem Cell-Conditioned Medium

- Anti-aging agent is a useful compound to prevent and repair aging process. Skin is the easiest tissue to be observed in aging process. Mesenchymal stem cell conditioned medium (MSC-CM) contains growth factors, cytokines, and tissue repair activity through paracrine signaling mechanism- The objective of this study was to observed the histological structure of skin aging model treated witt topical MSC-CM. Thirty-nine of 6 weeks old mice were divided into control group, aging gror.p, saline treated group and MSC-CM treated group. 1000 mg/kg BW of D-galactose were injected to induce skin aging. Mice were treated with saline and l0o/o MSC-CM cream in saline treated group and MSC-CM treated group, respectively. Skin samples were collected then processed by paraffin method- The samples were visualized by using Hematoxylin-eosin staining and Mallory Aniline Blue staining to evaluate histological structure and determined skin collagen density. Aging group showed that thc thickness of epidermis and dermis layers, the number of hair follicles, the number of blood vessels, and density of collagen fiber were decreased compared to the control group. On the other hand, MSCCM cream group showed that thickness of epidermis and dennis layers, the number of hair follicle, the number of blood vessels, and density of collagen fiber were increased compared to saline treated group. Based on histological study, MSC-CM cream has potential as tissue repair agent in skin aging.
