  • ABSTRACT One of a major constraint of soybean seed in storage is rapid deterioration of its quality, especially in the tropic. The experiment aimed to study physiology and biochemistry aspects of its deterioration during storage and to find method of seed storage for keeping the quality of soybean seed. A Randomized Completely Block Design was used in the experiment, with three factors. The first factor was moisture content, consisted of three levels i.e. 8%, 10%, dan 12%. The second was three kinds of bag, i.e. polyethylene plastic, wheat bag and aluminium foil. The third was storage period, consisted of seven levels, i.e. without storage, I, 2, 3, 4, 5 and 6 months. The results showed that the content of phospholipids of seed storaged at 8% moisture content did not significantly decrease until 6 months, while protein content of membrane, inorganic phosphor and specific activity of sucinate dehydrogenase decreased after 3 and 4 months. Specific activity of cytochrome oxidase and respiration rate of seed without storage at 12% moisture content and storaged in wheat bag is higher than the other treatments. The germination and vigor of seed storaged at 8% and 10% moisture content in all kinds of bags did not significantly decrease up to 6 months and higher as compared to the 12% moisture content. Key words: soybean, deterioration, physiology, biochemistry, storage INTISARI Deteriorasi benih kedelai selama penyimpanan menyebabkan kemerosotan mutunya khususnya di daerah tropis. Penelitian bertujuan untuk mempelajari aspek fisiologi dan biokimia deteriorasi benih kedelai selama penyimpanan dan menemukan cam simpan yang tepat untuk mempertahankan mutu benih kedelai tetap tinggi selama penyimpanan. Penelitian disusun dalam RCBD dengan 3 faktor. Faktor I adalah kadar air, terdiri atas 3 aras: 8%, 10%, 12%. Faktor II adalah kemasan, ada 3 jenis : kantong plastik polietilen, kantong gandum, kantong aluminium foil. Faktor III adalah lama simpan, terdiri atas 7 aras: tanpa disimpan, 1, 2, 3, 4, 5 dan 6 bulan. Hasa penelitian menunjukkan bahwa kadar fosfolipid benih yang disimpan pada kadar air 8% di dalam kantong aluminium foil belum menurun sampai akhir penyimpanan 6 bulan, kadar protein membran, fosfor anorganik dan aktivitas suksinat dehidrogenase menurun setelah 3 dan 4 bulan. Aktivitas spesifik sitokrom oksidase dan laju respirasi dari benih yang tanpa disimpan dengan kadar air 12% dan di dalam kantong terigu lebih tinggi dan berbeda nyata dengan lainnya.
