Pengaruh Vitamin C Terhadap Profil Farmakokinetika Teofilin Pada Sukarelawan Sehat

  • ABSTRACT Asthma bronchial and chronic obstructive pulmonary disease (COPD) are considerably common diseases in community. Theophylline, a bronchodilator which is relatively inexpensive, is used once until three times a day. Because of it's narrow therapeutic range, the dosage must be individualized in order to optimize the treatment and prevent side effects. Vitamin C is commonly used, because it has antioxidant properties while from several references vitamin C may increase the metabolism of drugs. Does vitamin C affect theophylline has not been clarified. Using randomized cross over design, volunteers were divided randomly into two groups, control and pretreatment group. The control group were given theophylline orally as a single dose 130 mg. Before starting the experiment, vitamin C 500 mg a day was given orally to pretreatment group for 7 consecutive days before of theophylline. In both groups after oral single dose of theophylline, the samples were collected serially for 24 hours to analyzed blood theophylline concentration using HPLC. Theophylline pharmacokinetic parameters were calculated with the assumption of one- compartement model using a standard method. The results shown that there were no significant changes in values of T max, C max, Ka, AUC 0-t and AUC 0-~, elimination half - life (tl /2), elimination rate constant (Kel) and Clearance (Cl) between control and pretreatment groups with p>0,05. In conclusion, pretreatment with vitamin C 500 mg daily for 7 days did not change the pharmacokinetic parameters of theophylline 130 mg single dose in the serum.
